Leveraging Metabolomics to Assess the Next Generation of Temozolomide-based Therapeutic Approaches for Glioblastomas
نویسندگان
چکیده
Glioblastoma multiforme (GBM) is the most common adult primary tumor of the central nervous system. The current standard of care for glioblastoma patients involves a combination of surgery, radiotherapy and chemotherapy with the alkylating agent temozolomide. Several mechanisms underlying the inherent and acquired temozolomide resistance have been identified and contribute to treatment failure. Early identification of temozolomide-resistant GBM patients and improvement of the therapeutic strategies available to treat this malignancy are of uttermost importance. This review initially looks at the molecular pathways underlying GBM formation and development with a particular emphasis placed on recent therapeutic advances made in the field. Our focus will next be directed toward the molecular mechanisms modulating temozolomide resistance in GBM patients and the strategies envisioned to circumvent this resistance. Finally, we highlight the diagnostic and prognostic value of metabolomics in cancers and assess its potential usefulness in improving the current standard of care for GBM patients.

متن کاملO-6: A Multi-Regional Study on New Approaches to Investigate The Quality of Human Sperm - Including DNA Fragmentation, Proteomics and Metabolomics
Background: Preliminary data has also shown that there is less fragmented sperm in 2nd and 3rd ejaculates compared to first ones which could be a major factor in determining the pregnancy outcome. Assessing this factor objectively and relating it to other parameters in sperm quality in this study could result in new prediction criteria for the pregnancy outcome. Materials and Methods: As one of...
متن کاملLoss of the mismatch repair protein MSH6 in human glioblastomas is associated with tumor progression during temozolomide treatment.
PURPOSE Glioblastomas are treated by surgical resection followed by radiotherapy [X-ray therapy (XRT)] and the alkylating chemotherapeutic agent temozolomide. Recently, inactivating mutations in the mismatch repair gene MSH6 were identified in two glioblastomas recurrent post-temozolomide. Because mismatch repair pathway inactivation is a known mediator of alkylator resistance in vitro, these f...
متن کاملEvaluation of the Anticancer Effect of Xanthium Strumarium Root Extract on Human Epithelial Ovarian Cancer Cells Using 1H NMR-Based Metabolomics
Epithelial Ovarian cancer is the leading cause of cancer mortality among women all over the world. As chemotherapeutics has many side effects, researchers have focused on the potential use of medicinal plants as natural antitumor agents. Xanthium strumarium studied in this work as an herbal anticancer agent. This study aimed to evaluate the antitumor effect and metabolic alterations ca...
متن کاملThe Changes in MGMT Promoter Methylation Status in Initial and Recurrent Glioblastomas.
To evaluate the mechanism of the development of therapeutic resistance after temozolomide treatment, we focused on changes in O(6)-methylguanine DNA methyltransferase (MGMT) and mismatch repair (MMR) between initial and recurrent glioblastomas. Tissue samples obtained from 24 paired histologically confirmed initial and recurrent adult glioblastoma patients who were initially treated with temozo...
